How It Works
Optimized for success
With XPack, SBI has developed an optimized N-terminal peptide sequence that targets the peptide—and any protein fused to it—to the inner surface of the exosome membrane. Our pre-built XPack-reporter constructs come in GFP, RFP, and luciferase options to cover a wide range of labeling needs.

Part 1: Packaging luciferase into exosomes with XPack
The XPack-Luciferase fusion protein is efficiently packaged as exosome cargo and delivered to target cells. HEK-293 cells were transfected with a plasmid encoding an N-terminal XPack-Luciferase fusion protein (Cat.# XPAK532PA-1). Forty-eight hours later, exosomes were isolated using ExoQuick-TC® and luciferase levels quantified and compared to exosomes isolated from untransfected cells (Figure 1). Only XPack-Luciferase exosomes show robust luminescence indicating efficient packaging of luciferase into exosomes.
Part 2: XPack-Luciferase Exosome Delivery to Target Cells
The XPack-Luciferase exosomes isolated as described above were then added to recipient HEK-293 cells to test for luciferase delivery. After 24-hours, robust luminescence from cells exposed to XPack-Luciferase exosomes shows delivery of the luciferase cargo to the target cells (Figure 2). In contrast, untreated cells or cells treated with exosomes from untransfected cells show no luminescence.
